Neuropsychiatric disorders including major depression, bipolar disorder, autistic spectrum disorders and schizophrenia affect a considerable segment of the population and thus may have a large-scale financial burden on the healthcare system and the society. In the same time, most psychiatric conditions still have unknown underlying causes and inefficient and/or suboptimal treatment.
In the past decade accumulating experimental and clinical data have suggested an important role of epigenetic dysfunction in the etiopathogenesis of neuropsychiatric diseases. Epigenetics, which deals with the heritable changes in gene expression that do not involve changes in DNA sequence, may provide a link between genetic and environmental factors, and neurobehavioral consequences.
It has been shown that epigenetic alterations at the levels of DNA methylation, histone modifications and noncoding RNAs can modulate transcriptional and translational events required for neurological processes. Recent reports demonstrate that epigenetic mechanismsare critically important in mediating precise gene regulation of neurogenesis, neural differentiation, plasticity and function.
Advances in our understanding of the epigenetic regulations of gene expression in the central nervous system and their role in neuropsychiatric disorders are highlighting novel pharmacological approaches with major therapeutic significancefor the treatment of these chronic, relapsing conditions. This book provides an overview of the major epigenetic mechanismsunderlying psychiatric dysfunctions and some modern approaches with the use of epigenome-modifying drugs in the field of neuropsychiatry.
